Abstract:AIM To obtain and sequence DNA clone of VP1unique region of human parvovirus B19from Chinese patients and to further study the genetic mutation of human par-vovirus B19of the Chinese strain.METHODS Two samples of VP1unique region of HPVB19were amplified from aplas-tic crisis serum of two Chinese children by PCR.The target fragment was linked with PUC19.Positive clones were cho-sen by identification of endonuclease cut and sequenced.RE┐SULTS A target fragment of480bp was amplified by PCR from the serum samples of one patient.Compared with the published B19VP1unique sequences,two base-pair and de-duced amino acid were found changed.CONCLUSION There are two mutations in the nucleotide sequence encoding the unique region of VP1of human parvovirus B19of the Chinese strain.
